Does anyone know the theoretical constraints on user numbers in MODX Revolution?

I can see two obvious constraints:

    • the database
    • the user interface in the Access Controls screen, which tries to show every user - this could perhaps be avoided by creating some other interface to the access controls

I'm looking at a system which could eventually have upwards of 3000 registered users. The site is actually relatively low-traffic, and most users will log in a maximum of once or twice per year. Perhaps 100 will log in once a week or more, and only half-a-dozen will log in each day. So the number of users looks like the biggest problem ...

No, modx.com has 40k+ users with no performance hit. You won't have any issues.
